چکیده

As reported in the previous paper by the authors, two kinds of precursor of plasminogen activator were presented by gel filtration method, which were named proactivator A and proactivator B.1) However the former fraction did not contain plasminogen, the latter on the other hand always coexisted with plasminogen, which seemed to be difficult to separate. Therefore, a question arises that fibrinolytic activity demonstrated by adding streptokinase (SK) to proactivator B fraction (with plasminogen) might not be mediated by the activation of proactivator but caused directly by the activation of plasminogen by SK. To resolve the question it is required that proactivator B can be obtained which was separated from plasminogen, and that fibrinolysis should be examined by using thus obtained proactivator B. The results in this paper are to prove that fibrinolytic activity demonstrated by proactivator B fraction is also mediated by an activator converted from proactivator B as in the case of proactivator A.

Purification and biochemical properties of human plasminogen.

Plasminogen is the plasma precursor of the fibrinolytic enzyme, plasmin. The activation of plasminogen isolated from various animal species by streptokinase, a protein of bacterial origin, has received considerable attention in recent years. A two-step reaction has been postulated wherein streptokmase reacts with a substance termed proactivator (1)) which is present in highest concentration in ...

The Fibrinolytic Pathway of Human Plasma

The conversion of the plasminogen proactivator to plasminogen activator by activated Hageman factor or its fragments has been recognized as an essential step in the conversion of plasminogen to plasmin. The plasminogen proactivator has been completely separated from prekallikrein and pre-PTA, two other proenzyme substrates of activated Hageman factor or its fragments. Plasminogen proactivator, ...

Gel Filtration of Fibrinolytic Components of Blood Plasma in Man and Rabbit.

This paper deals with a comparative aspect of the fibrinolytic system in man and rabbit, particularly relating to the "proactivator-like component" in rabbit plasma whose behavior is similar to proactivator A of human plasma fractionated by gel filtration and apparently acts as proactivator in the presence of a small amount of human globulin. The entity of this component has been remained yet q...

Hageman-factor-dependent fibrinolysis: generation of fibrinolytic activity by the interaction of human activated factor XI and plasminogen.

Human coagulation factor XI has been purified, and upon activation with Hageman factor fragments, was found to convert the fibrinolytic proenzyme plasminogen to plasmin. This proactivator activity was shown to be functionally and antigenically distinct from prekallikrein. When the gamma-globulin fractions of plasma deficient in Hageman factor, prekallikrein and factor XI were isolated, factor-X...
